Literature review
With age, stress and exposure to external factors, the function of granulosa cells that form the follicular environment becomes impaired.
This leads to impaired oocyte nutrition and is accompanied by the accumulation of DNA damage and chronic inflammation, as well as reduced resistance to oxidative stress.
One of the key regulators of these processes is the NAD⁺ molecule, which supports cellular repair, control of DNA damage and maintenance of stable functional activity of the follicle.
What is NAD⁺?
NAD⁺ (Nicotinamide Adenine Dinucleotide) is a universal molecule required for many biochemical reactions within the cell, primarily for maintaining the activity of the PARP and SIRT systems involved in DNA repair, anti-inflammatory activity and antioxidant protection.
DNA repair
PARP (Poly(ADP-ribose) polymerases): groups of enzymes that are activated in response to DNA damage in granulosa cells and initiate the DNA repair process.
When NAD⁺ levels are insufficient, the activity of this system is reduced, increasing the risk of granulosa cell apoptosis and consequently leading to arrest of oocyte development.
Anti-inflammatory activity
SIRT1,3,6 (sirtuins 1,3,6): a group of enzymes that control the activity of inflammatory mediators.
When NAD⁺ is deficient, sirtuin activity decreases, leading to increased inflammation and senescence* in follicular cells.
* Cellular senescence is a state in which a cell no longer divides while remaining metabolically active.
Antioxidant protection
SIRT1,2,3 (sirtuins 1,2,3): activators of enzymes responsible for the function of the cell's own antioxidant systems.
When NAD⁺ levels are insufficient, sirtuin activity decreases, leading to increased oxidative stress in follicular cells.

Quercetin is a natural flavonoid with antioxidant properties.
It may inhibit the activity of CD38, an enzyme that actively consumes NAD⁺ and is one of the factors contributing to the age-related decline in intracellular NAD⁺ levels.
Thus, quercetin may help preserve the cell's own NAD⁺ reserves.
Betaine is a methyl-group donor involved in methylation processes and homocysteine metabolism.
Its inclusion in the complex complements the action of the NAD⁺ precursor and supports metabolic processes associated with its utilisation
